Paraffin Tissue Block Preparation for both High Volume and Low Volume Histology Laboratories
The new PELCO Embedding Studio™ introduces features that improve efficiency and functionality over existing embedding centers on the market, meeting the needs for both high and low volume histology laboratories. It is built with the latest innovations on top of trusted technology. The addition of the PELCO Freezer™ module allows partially solidified Super Mega blocks as well as standard size blocks to be moved off of the PELCO Chiller™ plate into the PELCO Freezer™ to complete polymerization and free up the chiller for more blocks. This clears the PELCO Chiller™ plate to allow continued embedding, greatly increasing throughput. The PELCO Freezer™ is a both holding chamber for all tissue blocks as well as a place to complete polymerization at the optimal temperature to solidify paraffin. Thermoelectric cooling is used, eliminating the use of refrigerant gases. This meets new green regulatory requirements coming into force in various countries.
The PELCO Embedding Studio™ enhances tissue embedding workflow and is specifically designed to accommodate the embedding of large super mega tissue blocks. The PELCO Freezer™ is unique in histology, bringing a workflow enhancement that no other tissue embedding center provides.
The PELCO Dispenser™ dispenses paraffin and provides paraffin management. It is also the control module which contains the control center for the PELCO Embedding Studio™ as a whole. At the top right side of the module is an easy to program touchscreen control panel for setting days of the week, start time, stop time, and temperature.
The 3.8-Liter paraffin reservoir is filled through the top access lid. The dispensing arm has four heated forceps holders on each side with a precise paraffin flow control knob for the dispensing nozzle. The PELCO Dispenser™ has a large heated work surface (ranging from 50°C to 75°C) to accommodate warmed embedding molds and tissue cassettes ready for embedding. This includes vertical trimming plates on both forceps holder/warmers and two horizontal trimming plates on the heated work surface. It also includes a 61 x 46mm cold spot (ranging from -5°C to 5°C) for tissue orientation with any size of embedding mold.
Waste paraffin is collected in two heated paraffin collection trays. These silicone rubber trays are heated to prevent the buildup of polymerized paraffin and allows easy removal of the waste paraffin.

The PELCO Warmer™ module will hold 800+ standard embedding molds or up to 100 Super Mega molds. The door of the warming chamber folds back for efficient access. The base has a large warming tray with a sliding cover, which accommodates most tissue processing baskets. The PELCO Warmer™ has a temperature range of 45°C to 70°C.

The PELCO Chiller™ cold plate module may be operated independently or connected and controlled by the PELCO Embedding Studio™ control program.
The PELCO Chiller™ is cooled by thermoelectric cooling technology with a temperature range of -5°C to 5°C. It accommodates up to 50 standard size embedding molds or 12 Super Mega embedding molds.
A Transfer Tray may be used on the PELCO Chiller™ plate. Each Transfer Tray holds up to 21 embedding molds or 6 Super Mega Molds. When filled the Tray may be quickly transferred to the PELCO Freezer™ box to allow continuous embedding. This is especially important if embedding Super Mega tissue blocks as they may take 20 to 35 minutes to complete polymerization. The PELCO Chiller™ plate accommodates up to 2 Transfer Trays.

The PELCO Freezer™ is the fourth module for the PELCO Embedding Studio™. It may be used alongside the other three modules or on its own. The PELCO Freezer™ operates at a temperature range of -5°C to 5°C, mirroring the operating temperature of the PELCO Chiller™ cold plate module. This is the optimal temperature range for paraffin polymerization as colder temperatures may cause paraffin blocks to crack.
The PELCO Freezer™ utilizes thermoelectric cooling (TEC) technology rather than freon gas compressors used in most refrigeration systems. It has five shelves and each shelf accommodates two Transfer Trays. It has a capacity of up to 210 standard paraffin embedded tissue blocks or up to 60 Super Mega paraffin embedded tissue blocks.
Because tissue blocks may be transferred to the PELCO Freezer™ to complete polymerization, the PELCO Chiller™ will always be available for more. This is particularly helpful with Super Mega tissue blocks that require a long polymerization time. These tissue blocks may be stored in the PELCO Freezer™ until ready to pick up for sectioning at a microtome station. The large viewing window allows good visibility for whether tissue blocks are available and ready for sectioning without opening the door of the PELCO Freezer™.

The highly-durable Paraffin Scraper can be used to remove paraffin wax from the PELCO Embedding Studio™ and work station surfaces.

Tissue processing and embedding medium (paraffin) composed of light blue pellets. ParaPro™ Blue may be used with all tissues, but is ideal to use with small biopsy tissue specimens for better visualization of the tissue. Optimized for dermatology and urology specimens. A blend of highly refined paraffin with additives of synthetic polymers. Section down to 3µm thickness with excellent ribbon continuity; melting point 56°C.

Tissue embedding medium for faster penetration. Paraplast Plus® is recommended for large tissues and tissues which are difficult to process. Section down to 2µm thickness with excellent ribbon continuity; melts rapidly at 56-67°C. Double filtered paraffin containing plastic polymers of regulated molecular weights and a small percent (0.8%) of dimethyl sulphoxide (DMSO) for more efficient infiltration of dense and difficult to process tissues. No stainable residue.

Optimum tissue embedding medium composed of clear pellets of low-molecular weight polymers and highly purified paraffin. A low viscosity paraffin. Low melting point of 52° to 54°C. Recommended for low temperature processing, small biopsy tissues and for general tissue embedding. Excellent sectioning qualities. Sections down to 2µm thickness with exceptional compression resistance and ribbon continuity. Cut sections are free of distortion; sample morphology is preserved. Low viscosity permits complete infiltration of dense tissue. Sections adhere perfectly to slides without leaving stainable residue.

27197-1, 27197-2 and 27197-4 have higher sides than competitor super mega base molds. The higher sides allow for more paraffin to be used during embedding assuring a stronger bond of the paraffin block to the cassette.

PolyShield® is a liquid paraffin repellent used as a protective coating on laboratory surfaces that are exposed to paraffin residue. PolyShield® can also be used to clean paraffin that has adhered to lab countertops and equipment.
