KODAK Electron Microscope Film 4489

KODAK Electron Microscope Film 4489 has been reformulated. This new product is based on the "state-of-the-art" T-grain technology that has been requested by customers. It is the highest quality, high contrast TEM film available in today's market place for transmission electron microscope imaging of biological samples.

The improved reformulated KODAK Electron Microscope Film 4489 offers:

Improved Uniformity: Even development across the entire film plane. You can use nitrogen burst developer agitation; 2 second bursts every 8 seconds. If hand agitation is used, insert film rack quickly into the developer tank and tap rack on the tank bottom sharply for 5 seconds to dislodge any air bells on the film surface. Agitate film in developer tank for 5 seconds every 30 seconds in a side-to-side and back and forth direction in the developer tank.

Increased Speed: Slightly faster than the previous 4489 product. Make an exposure series test to determine an exposure density that is best suited for your work. After processing, evaluate the negatives so you will be able to calibrate the instrument exposure setting to obtain a negative density suited specifically to match your needs.

Processing chemicals and development times have not changed but you should confirm your film exposure and processing procedures whenever using a new or reformulated film.

The new film will begin to enter the ordering system in due time. A blue label marked "New and Improved" on the box will notify you that you have received the reformulated film.

4489 Features:

  • 7 mil (0.007)(0.18mm) ESTAR thick base
  • Extremely fine grain
  • Blue-sensitive emulsion
  • Notched in right-hand side of the top edge of the film sheet, emulsion up
  • Emulsion overcoat (0.5 to 1.0µm thick) or hardened gelatin which reduces susceptibility to abrasion
  • Clear gelatin layer on back of base to reduce curl

Recommended for use in electron microscopy and electron diffraction in the medium electron-energy range (40-100kV). The base material is tear-resistant and dimensionally stable. This polyester base contains no solvents, thus eliminating one possible source of contamination in the column of the EM. All film faces the same direction in its package.

Storage Keep unopened film packages in a cool, dry place. A temperature of 21°C (70°F) or lower and a relative humidity of 50% are recommended. For long periods of storage, keep the film at a temperature of 4.5°C (40°F) or lower.

Avoid Long Prepump Times (see PELCO Vacuum Desiccator). Moisture condensation must be avoided for film that has been refrigerated. Allow unopened film package to stand at room temperature at least 2 hours before use.

Handle film by edges only and avoid accidental contact of either film surface with film holders, film racks, processing hangers or other objects.

High concentrations of dust particles in the area of the film causes "pinholes" in processed negatives and black spots on the photographic film.

Handling film during periods of 25% or lower relative humidity can cause static charge build-up on the film. This attracts dust particles, and the static charge can discharge abruptly causing unwanted exposure on the emulsion. Prior to exposure, static build-up can be minimized by wiping the top of the workbench with a damp cloth before loading film into holders (removing dust and increasing humidity in the immediate area). After exposure in the vacuum of the EM, the film is extremely dry. Although the film reaches equilibrium very quickly with the air humidity, it is a good idea to avoid sudden movements of the film that might cause friction and possible static discharge.

Safelight Filters KODAK OA (greenish yellow), 1A (light red), OC (light amber), with a 15 watt bulb. Keep the safelight lamp at least 1.2m (4 ft.) away from the film-handling area.

Developer KODAK D-19 (1 part developer to 2 parts water) at 20°C (68°F) for 4 minutes. Development times longer than 4 minutes will result in increased background fog. Agitate manually or by nitrogen-burst. Replace the developer after 1 week or after processing the equivalent of 120 - 1-1/4 x 4 inch sheets per gallon.

Rinse in running water for a full 1-1/2 minutes at 18 to 21°C (65 - 70°F). Agitate.

Fix at 18 to 21°C (65 - 70°F) for 8 - 10 minutes in KODAK Fixer. Agitate frequently.

Wash the film 20 to 30 minutes in running water at 18 to 21°C (65 - 70°F). KODAK Hypo Clearing Agent can be used to reduce washing time and to conserve water.

To minimize drying marks, treat the film in KODAK PHOTO-FLO solution after washing or wipe surfaces carefully with chamois or a soft viscose sponge. Dry the film in a dust-free place (see Table-top Film Dryer).

KODAK Electron Image Film SO-163

  • Twice as fast as 4489
  • Broad exposure and development range
  • 7 mil (0.007")(0.18mm) ESTAR thick base
  • Extremely fine grain
  • Blue-sensitive emulsion
  • Notched in right-hand side of the top edge of the film sheet, emulsion up
  • Emulsion overcoat (0.5 to 1.0µm thick) of hardened gelatin which reduces susceptibility to abrasion
  • Clear gelatin layer on back of base to reduce curl

SO-163 will show a tendency to curl, and requires a longer pump-down time (see PELCO® Vacuum Desiccator). When using SO-163 for the first time, recalibrating the microscope exposure device is recommended.
